Preparation

Dissolve NaNO₃ → K₂HPO₄ → MgSO₄·7H₂O → NaHCO₃ → H₃BO₃ → 1 mL of the trace element solution → CaCl₂·2H₂O → adjust the pH → Na₂SiO₃·9H₂O last → autoclave → add 0.5 mL of the vitamin solution aseptically.

Notes

  • MLA was developed as a single medium for a wide range of freshwater phytoplankton — convenient when one collection holds several different taxonomic groups.
  • The primary stock solutions for the MLA trace element solution are the same as for f/2; in addition, a primary selenite stock solution is prepared (Na₂SeO₃ 12.9 mg per 100 mL).
  • Na₂SiO₃·9H₂O is added only when diatoms are being cultured.
  • The composition is identical to the f/2 working solution; the only difference is sodium selenite. Primary stock solution No. 6: Na₂SeO₃ 12.9 mg per 100 mL. Selenium is required by dinoflagellates and by some golden algae; it is omitted for green algae and cyanobacteria.
  • The five solutions are prepared and stored in separate, clearly labelled bottles. They must not be mixed with one another before being added to the working solution.
  • Vitamins are NOT autoclaved — sterile filtration through a 0.22 µm membrane only. Thiamine loses 40–70 % of its activity at 121 °C and degrades at pH above 8 even without heating, so vitamins are added to the cooled medium immediately before inoculation. Refreezing of aliquots is not permitted.
  • Biotin is poorly soluble in cold water — dissolve it in 1 mL of 50 % ethanol or in warm water at pH 8, then make up the volume. The B₁₂ solution has a characteristic pink-red colour; loss of colour means the vitamin has decomposed.
